Luke Macfarlane was born on 19 January 1980 in London, Ontario, to Thomas and Penny Macfarlane. Thomas Macfarlane was the Director of Student Health Services at the University of Western Ontario, and Penny worked as a mental health nurse at a hospital in London, Ontario.
Macfarlane has two siblings: a twin sister named Ruth Macfarlane and an older sister, Rebecca Macfarlane. The siblings attended London Central Secondary School before Macfarlane enrolled at the Lester B. Pearson School for the Arts and the Julliard School in New York City, where he studied Drama.
